Abstract:
Using the seeds and stem segments of
Dendrobium officinale as raw materials, the effects of medium and plant growth regulators on protocorm induction and proliferation were studied by single factor experiment. The results showed that adding a certain concentration of auxin and cytokinin could effectively promote protocorm induction of
Dendrobium officinale, but too high or too low concentration was not conducive to the induction and proliferation of the original bulb. The induction rate of protocorm with seed as explant was higher than that with stem as explant; the most suitable medium for protocorm induction of seed explant was MS+NAA 0.1 mg·L
-1+agar 7.0 g·L
-1+sucrose 30.0 g·L
-1 with the induction rate of 98.7%, and the most suitable medium for protocorm proliferation was MS+NAA 0.1 mg·L
-1+6-BA 0.5 mg·L
-1+agar 7.0 g·L
-1+sucrose 30.0 g·L
-1.
