Abstract:
In order to optimize the sterilization method of the seeds of Mucuna plants, the best sterilization method of the seeds of Mucuna plants was screened to provide the basis for the establishment of the tissue culture rapid propagation technology. Taking the seeds of four kinds of Mucuna plants as explants and the soaking time of alcohol, sodium hypochlorite and mercury bichloride as three factors, the best sterilization method of Mucuna explants was selected through the orthogonal test of three factors and three levels. The results showed that the optimal treatment for the sterilization of Mucuna calophylla was 60 s of alcohol followed by 10 min of sodium hypochlorite and then 10-15 min of mercury bichloride. The optimal treatment for the sterilization of Mucuna birdwoodiana Tutcher was 60 s of alcohol followed by 10-15 min of sodium hypochlorite and then 15 min of mercury bichloride. The optimal combination for the low pollution rate of Mucuna macrocarpa was sodium hypochlorite for 15 min after 40 s of alcohol and then mercury bichloride for 10 min, while the optimal combination for the high germination number was sodium hypochlorite for 10 min after 60 s of alcohol and then mercury bichloride for 15 min. The optimal combination for the low pollution rate of Mucuna sempervirens was sodium hypochlorite for 15 min after 40 s of alcohol and then mercury bichloride for 10 min, while the optimal combination for the high germination number was sodium hypochlorite for 10 min after 60 s of alcohol and then mercury bichloride for 10 min. The results showed that the optimal combinations of seed sterilization were different in different species of Mucuna plants.
