1株灰黄霉素产生菌的诱变选育及其发酵条件优化

    Mutation Breeding of a Griseofulin-producing Strain and the Optimization of Its Fermentation Conditions

    • 摘要: 以灰黄霉素产生菌ji-2为出发菌株经过紫外线与氯化锂诱变处理,选育出灰黄霉素效价高且发酵周期较短的菌株UL100-80,其效价为91 304 μg·mL-1,比出发菌株ji-2提高了47.8%,通过连续传代培养,其遗传稳定性良好。对UL 100-80发酵条件进行优化,优化后液体发酵培养基:大米粉150 g·L-1,NaCl 8.0 g·L-1,CaCO3 8.0 g·L-1,黄豆饼粉1.0 g·L-1,KH2PO4 6.0 g·L-1,KCl 7.0 g·L-1,NaNO3 1.0 g·L-1,FeSO4 1.0 g·L-1,pH自然;优化后的培养条件:培养40 h的种子液以12%的接种量接种,28℃下培养12 d,灰黄霉素的效价从未优化前的91 304 μg·mL-1提高到124 965 μg·mL-1,提高了36.9%,与出发菌株ji-2相比,UL100-80产灰黄霉素的效价提高了1.02倍,发酵时间上缩短了48 h。

       

      Abstract: By taking the griseofulin-producing strain ji-2 as the original strain, the strain UL100-80 with high potency of griseofulvin and short fermentation period was selected after the mutagenesis with ultraviolet ray and lithium chloride. The potency of the strain UL100-80 was 91 304 μg·mL-1,which was 47.8% higher than that of the original strain ji-2, and the genetic stability was good through the continuous subculture. The fermentation conditions of UL100-80 were optimized, and the optimized liquid fermentation medium was as follows:rice flour 150 g·L-1, NaCl 8.0 g·L-1, CaCO3 8.0 g·L-1, soybean powder 1.0 g·L-1, KH2PO4 6.0 g·L-1, KCl 7.0 g·L-1, NaNO3 1.0 g·L-1, FeSO4 1.0 g·L-1, and pH was natural. The optimized culture conditions were as follows:the seed solution cultured for 40 h was inoculated at 12% dose, and cultured at 28℃ for 12 d. The potency of griseofulvin was increased from 91 304 μg·mL-1 before the optimization to 124 965 μg·mL-1, with an increase of 36.9%. While compared with the original strain ji-2, the potency of griseofulvin produced by UL100-80 increased by 1.02 times, and the fermentation time was shortened by 48 h.
