Abstract:
In this study, by using two callus cell lines of
Vitis davidii as the materials, the total antioxidant activities of different callus cell cultures of
Vitis davidii were determined by using the colorimetric method and spectrophotometric method, thus to analyze the effects of different culture stages and methods such as cinnamic acid, light conditions, dark conditions, temperature (4℃, 37℃), chitosan and ultraviolet-B (UV-B) on the total antioxidant activity of
Vitis davidii callus. The results showed that the anthocyanin and non-anthocyanin polyphenol extracts from the red callus of
Vitis davidii at different culture stages had strong antioxidant activities. The antioxidant activity of anthocyanin extracts from the red callus of
Vitis davidii at different culture stages showed double peaks, with the maximum value of 7.43 U·mg
-1 (FW), and that of non-anthocyanin polyphenol extracts was up to 28.36 U·mg
-1 (FW). However,the antioxidant activities of anthocyanin and non-anthocyanin polyphenol extracts from light yellow-green callus at different culture stages were very low. The total antioxidant activities of anthocyanin and non-anthocyanin polyphenol extracts from the callus of
Vitis davidii under different treatments had abundant variations. The treatment with temperature of 4℃ was the most effective in improving the antioxidant activity of anthocyanin extracts from the red callus of
Vitis davidii, while the treatment with chitosan was the most effective in improving the total antioxidant activity of non-anthocyanin polyphenol extracts. The antioxidant activities of anthocyanin and non-anthocyanin polyphenol extracts from light yellow-green callus were extremely low, and the treatments with cinnamic acid, illumination condition, temperature, chitosan and UVB had little effect on their antioxidant activities.
