摘要: 竞争性等位基因特异性PCR(KompetitiveAllele-SpecificPCR,KASP)是一种基于荧光的同质基因分型技术,可对基因组的单核苷酸多态性位点以及插入/缺失多态性进行双等位基因分型检测,具有检测快速,成本低,准确率高等优点。阐述了KASP检测体系组成、扩增原理与反应步骤,以及相对于传统SNP检测方法的优势,总结其目前在动物遗传鉴定、功能基因分析、分子标记选择(MAS)辅助育种中的研究进展,以期为KASP技术在动物领域的应用提供参考依据。Abstract: Kompetitive allele-specific PCR (KASP) was a fluorescence-based homogeneous genotyping technique, which could perform the biallelic genotyping detection on the single nucleotide polymorphism sites and insertion/deletion polymorphisms in the genome. It had the advantages of rapid detection, low cost and high accuracy. The composition, amplification principle and reaction steps of KASP detection system were described in this paper, as well as its advantages over the traditional SNP detection methods. Then, the current research progresses in animal genetic identification, functional gene analysis and molecular marker-assisted selection (MAS) assisted breeding were summarized, in order to provide reference for the application of KASP technology in animal field.
