利用基因编辑技术创制福香占糯稻新种质

    Creation of Novel Glutinous Rice Germplasm of Fuxiangzhan(FXZ) Using Genome Editing

    • 摘要: 福香占是新育成的优质、抗稻瘟病、长粒籼稻品种,具有较好的推广潜力。本研究旨在利用基因编辑技术快速创制福香占糯稻材料。通过筛选和优化靶点序列,成功地利用CRISPR/Cas9技术敲除水稻颗粒结合型淀粉合酶基因(Waxy, Wx),获得了Wx基因的缺失突变体。通过后代农艺性状和米质鉴定,结果表明敲除材料的淀粉含量从16.3%下降至1.67%~1.81%,达到了糯稻水平。此外,敲除植株在株叶态、产量和抗性方面并没有显著变化。此项研究为福香占的应用推广提供了新的途径,并丰富了抗瘟糯稻的种质资源。

       

      Abstract: Gene editing is an emerging genetic improvement technology that allows for rapid and precise modification of crop traits. FXZ is a newly developed high-quality, blast-resistant, long-grain indica rice variety with great potential for commercialization. This study aimed to utilize gene editing technology to create FXZ glutinous rice materials rapidly. By screening and optimizing target sequences, the rice granule-bound starch synthase gene (Waxy, Wx) was successfully knocked out using CRISPR/Cas9 technology, resulting in Wx gene knockout mutants. Through progeny agronomic traits evaluation and amylose content determination, the results showed that the amylose content of the knockout materials decreased from 16.3% to approximately 1.67%-1.81%, meeting the criteriaof glutinous rice. Additionally, there were no significant changes observed in plant morphology, yield, and resistance in the mutant plants. This study provides a new approach for the application and promotion of FXZ and enriches the genetic resources of blast-resistant glutinous rice.
