    毛竹笋木聚糖提取工艺优化及其抗氧化活性分析

    Optimization of Xylan extraction process and its antioxidant activity in bamboo shoot

    • 摘要: 为了实现竹笋加工废弃物的资源转化与利用,采用碱提醇沉法提取毛竹笋中的木聚糖,通过单因素试验和正交试验优化提取工艺,利用高效液相色谱法和傅里叶变换红外光谱分析毛竹笋木聚糖的理化性质,并通过DPPH法和ABTS法评价毛竹笋木聚糖的体外抗氧化活性。结果表明:在提取时间3.5 h、碱液质量浓度0.1 g·mL−1、料液比1∶25,提取温度100℃的条件下,毛竹笋木聚糖提取率最高,可达20.86%。单糖组成和FT-IR分析表明木糖可能是构成毛竹笋木聚糖的主要成分。体外抗氧化性试验表明毛竹笋木聚糖具有一定的抗氧化活性,当浓度为8 mg·mL−1时,其对DPPH自由基的清除率达到94.75%,IC50值为1.0 mg·mL−1;当浓度为20 mg·mL−1时,其对ABTS自由基的清除率为93.09%,IC50值为3.85 mg·mL−1

       

      Abstract: In order to realize the resource transformation and utilization of bamboo shoot processing waste, this study used alkaline extraction of xylan from bamboo shoots, optimized the extraction process by one-way and orthogonal tests, analyzed the physicochemical properties of xylan from bamboo shoots by high performance liquid chromatography and Fourier transform infrared spectroscopy, and evaluated the in vitro antioxidant activity of xylan from bamboo shoots by DPPH method and ABTS method.The results showed that the highest extraction rate of 20.86% was achieved at an extraction time of 3.5 h, an alkali mass concentration of 0.1 g·mL−1, a liquid-to-feed ratio of 1∶25, and an extraction temperature of 100 ℃. The monosaccharide composition and FT-IR analysis showed that xylose may be the main structure of xylan in bamboo shoots. The xylan of bamboo shoots has a certain antioxidant activity, the scavenging rate of DPPH free radicals reached 94.75% at a concentration of 8 mg·mL−1, with an IC50 value of 1.0 mg·mL−1, and the scavenging rate of ABTS free radicals was 93.09% at a concentration of 20 mg·mL−1, with an IC50 value of 3.85 mg·mL−1.
