大肠杆菌过表达WcaB对氨基糖苷类抗生素敏感性的影响

    Effect on the Sensitivity of Escherichia Coli Overexpressing WcaB to Aminoglycoside Antibiotics

    • 摘要: 为寻找新的靶点以加速解决由于抗生素滥用而引发的细菌耐药问题,通过利用pCA24N表达载体构建大肠杆菌BW25113过表达乙酰基转移酶WcaB菌株,在对数期进行氨基糖苷类抗生素杀伤效果测试,并进一步测定大肠杆菌BW25113过表达WcaB对氨基糖苷类抗生素的最小抑菌浓度(MIC)和生长曲线。结果表明:WcaB过表达后会抑制细菌生长,氨基糖苷类抗生素对WcaB过表达株的MIC没有明显变化,WcaB过表达使细菌在庆大霉素或卡那霉素处理下的存活率降低,证明WcaB过表达提高了大肠杆菌对氨基糖苷类抗生素的敏感性。研究结果显示乙酰基转移酶在增强抗生素对大肠杆菌产敏感性方面具有重要的作用,该酶有望成为大肠杆菌新的药理靶点。

       

      Abstract: In order to find the new targets to accelerate the solution of bacterial resistance problem caused by the antibiotic abuse, Escherichia coli BW25113 overexpressing acetyltransferase WcaB strain was constructed by using the pCA24N expression vector, and the killing effect of aminoglycoside antibiotics was tested in the logarithmic phase. Then, the minimum inhibitory concentration (MIC) and growth curve of Escherichia coli BW25113 overexpressing WcaB to aminoglycoside antibiotics were further determined. The results showed that: the overexpression of WcaB would inhibit the bacterial growth, and the MIC of aminoglycoside antibiotics to WcaB overexpressing strain did not change significantly. The overexpression of WcaB reduced the survival rate of bacteria treated with gentamicin or kanamycin, indicating that the overexpression of WcaB increased the sensitivity of Escherichia coli to aminoglycoside antibiotics. The results showed that acetyltransferase played an important role in enhancing the sensitivity of antibiotics to Escherichia coli, and the enzyme was expected to become a new pharmacological target for Escherichia coli.
