基于16S rDNA序列的秋茄内生细菌多样性分析

    Diversity analysis of endophytic bacteria in Kandelia candel based on 16S rDNA sequence

    • 摘要: 分析闽江河口湿地秋茄根、枝和叶中可培养内生细菌的多样性,为秋茄内生细菌群落结构、分布、稳定性以及功能内生细菌的研究提供支撑。从秋茄Kandelia candel不同组织中分离可培养内生细菌,基于16S rDNA基因序列比对和系统发育分析内生细菌多样性。结果表明,从秋茄3个组织中分离出36株可培养内生细菌,分属于11属22种(其中2株未被正式命名),其中芽孢杆菌属Bacillus和弧菌属Vibrio为优势菌属,分别占总株数的21.2%和19.4%。不同组织中内生细菌的分布和多样性存在差异,根中内生细菌数量最多(27株,占总株数的75%),枝次之(6株,占总株数的16.7%),叶最少(3株,占总株数的8.3%)。秋茄内生细菌具有丰富的物种多样性,不同组织之间呈现出差异性,根中内生细菌多样性为最丰富。

       

      Abstract: To study the diversity of culturable endophytic bacteria in the roots, branches and leaves of Kandelia candel in the wetland of Minjiang River estuary, and to provide support for the study of community structure, distribution, stability and functional endophytic bacteria of Kandelia candel. In this study, culturable endophytic bacteria were isolated from different tissues of Kandelia candel, and the endophytic bacterial diversity was analyzed based on 16S rDNA gene sequence alignment and phylogeny. The results showed that 36 strains of culturable endophytic bacteria were isolated from three tissues of Kandelia candel, which belongs to 11 genera and 22 species(2 strains among which are not officially named). Bacillus and Vibrio were the dominant strains, accounting for 21.2% and 19.4% of the total strains, respectively. There were differences in the distribution and diversity of endophytic bacteria in different tissues, among which the number of endophytic bacteria was the highest in roots(27 strains, 75% of the total number of strains), followed by branches(6 strains, 16.7% of the total number of strains), and the least in leaves(3 strains, 8.3% of the total number of strains). The endophytic bacteria of Kandelia candel have rich species diversity in the wetland of Minjiang River estuary, and the diversity of endophytic bacteria in the root is the most abundant.
