大肠杆菌lysR基因敲除和表达对氨基糖苷类抗生素耐受性分析

    • 摘要: 为了解决抗生素滥用造成的细菌耐药问题,寻找抗生素作用的新靶点,检测了野生型菌株BW25113(WT)、lysR基因敲除菌株(ΔlysR)对氨基糖苷类抗生素(庆大霉素、妥布霉素、链霉素)、喹诺酮类抗生素、β-内酰胺类抗生素的杀菌表型差异,并进一步测试庆大霉素对过表达菌株BW25113-pCA24N和BW25113-pCA24N(lysR)的杀菌表型,最后通过外源添加赖氨酸检测庆大霉素对野生型菌株BW25113(WT)、lysR基因敲除菌株(ΔlysR)的杀菌表型,以此确定lysR基因对细菌耐药的重要性。结果表明:与野生型菌株BW25113(WT)相比,lysR基因敲除菌株(ΔlysR)对氨基糖苷类抗生素具有耐受性,对喹诺酮类抗生素、β-内酰胺类抗生素不具有耐受性,且过表达菌株BW25113-pCA24N(lysR)可以恢复对庆大霉素的敏感性,外源添加赖氨酸可以使lysR基因敲除菌株(ΔlysR)对庆大霉素的耐受表型得到缓解。以上结果证实了lysR基因对大肠杆菌耐受氨基糖苷类有显著影响,可能存在独特的耐药机制可作为新的抗生素作用靶点。

       

      Abstract: In order to solve the problem of bacterial resistance caused by the abuse of antibiotics and find new targets for the action of antibiotics, In this study, the bactericidal phenotypes of wild-type strain BW25113(WT)and lysR gene knockout strain (ΔlysR)against aminoglycoside antibiotics(gentamicin, tobramycin, streptomycin), quinolones and β-lactam antibiotics were detected. Furthermore, the effects of gentamicin on the overexpression strain Bw25113-pca24N and BW25113-pca24N were further tested The bactericidal phenotype of BW25113-pca24N(lysR)was determined. Finally, the bactericidal phenotype of gentamicin on the wild type strain BW25113(WT)and lysR gene knockout strain(ΔlysR)was detected by exogenous addition of lysine to determine the importance of lysR gene in bacterial resistance. The results showed that: Compared with the wild type strain BW25113(WT), the lysR gene knockout strain(ΔlysR)was resistant to aminoglycoside antibiotics, but not to quinolones and β-lactam antibiotics, and the overexpression strain BW25113-pca24N(lysR)could restore the sensitivity to gentamicin. Exogenous addition of lysine could alleviate the gentamicin resistance phenotype of the lysR gene knockout strain(ΔlysR). The lysR gene has a significant effect on aminoglycosides resistance in E. coli, and there may be a unique resistance mechanism of lysR gene, which can be used as a new target of antibiotics.
