云南金花茶叶片愈伤组织诱导研究

    Study on Callus Induction from Leaf Explants of Camellia fascicularis

    • 摘要: 为筛选云南金花茶叶片愈伤组织诱导培养基配方,以云南金花茶组培苗的叶片为材料,探讨激素、光照、放置方式对叶片愈伤组织诱导的影响,通过正交试验优化愈伤组织诱导培养基配方。结果表明:叶片背面与培养基接触、全程暗培养的方式最有利于云南金花茶叶片愈伤组织的诱导,叶片愈伤组织诱导最佳培养基配方为1/2MS + 45 g·L−1 蔗糖 + 3.0 mg·L−1 NAA。研究建立了云南金花茶叶片愈伤组织诱导技术体系,为下一步开发愈伤组织遗传转化体系奠定了前期基础。

       

      Abstract: To screen the optimal medium formulation for callus induction from leaves of Camellia fascicularis, leaves from tissue-cultured seedlings were used as materials. The effects of hormones, light conditions, explant orientation, and medium composition on leaf callus induction were investigated, and the callus induction medium formulation was optimized through orthogonal experiments. The results showed that placing the leaf abaxial side in contact with the medium and maintaining complete darkness during cultivation were most conducive to callus induction. The optimal medium formulation for leaf callus induction was 1/2MS + 45 g·L−1 sucrose + 3.0 mg·L−1 NAA. This study established a technical system for callus induction from leaves of C. fascicularis, laying a preliminary foundation for the subsequent development of a genetic transformation system using callus.
