鲜食木薯华南9号组培快繁体系的建立

    Establishment of tissue culture rapid propagation system of cassava cultivar SC9

    • 摘要: 为建立木薯华南9号组培快繁技术体系,以鲜食木薯华南9号顶芽和带腋芽茎段作为外植体,对MS基本培养基进行改良,设计了自创基本培养基ZCM,研究培养基中添加不同浓度的6-BA和IBA对增殖、生根的影响,并探讨炼苗方式对移栽成活率的作用。结果表明:外植体在MS和ZCM培养基上培养均可诱导萌芽和生长,茎段和顶芽无菌苗诱导率分别为66.7%和46.7%,外植体采用带腋芽茎段为佳。继代增殖培养的适宜培养基是MS和ZCM,增殖系数分别为3.5和3.4。适宜的生根培养基是MS和ZCM,生根率达到100%,根数多且长。组培生根苗采用先出瓶后液培炼苗方法,可有效提高移栽成活率。

       

      Abstract: This study aims to develop a rapid propagation method for the cassava cultivar ‘Huanan 9’ using an apical bud and stem segment with one axillary bud as explants. The MS basal culture medium was modified,and a self-created basal culture medium(named ZCM)was designed to conduct experiments in parallel with the MS medium.Cultivate and induce sterile seedlings in MS and ZCM media, thus different concentrations 6-BA and IBA were added to MS or ZCM medium to study the effects of different concentration ratios of plant-growth regulator on the subculture proliferation, and different concentrations NAA and IBA were added to MS or ZCM medium to study the effects of rooting. Screening suitable proliferation and rooting medium formulas.And explore the effect of seedling hardening methods on the survival rate of tissue cultured rooting seedlings after transplantation.The results indicate that both MS and ZCM media can effectively induce germination and growth of the plants, achieving a sterile seedling induction rate of 66.7% for stem segment and 46.7% for apical bud,stem segments with one axillary bud are better explants.The optimal medium for proliferation culture was identified as MS or ZCM,the proliferation coefficients are 3.5 and 3.4, respectively. MS or ZCM were proved suitable for rooting,achieving a rooting rate of 100%.The tissue-cultured seedlings developed numerous and long roots. After removal from the tissue culture bottle, the rooted seedlings were cultured with a nutrient solution outside the bottle, which improved the survival rate during transplantation.
