多花黄精组培快繁技术优化

    Optimization of Tissue Culture and Rapid Propagation Technology for Polygonatum cyrtonema Hua

    • 摘要: 为优化多花黄精组培快繁技术体系,以多花黄精7分成熟浆果、成熟种子、3年生苗的地下块茎作为试验材料,筛选最佳的外植体,进一步通过试验筛选最佳外植体的诱导培养基、芽苗增殖继代培养基及生根培养基的最佳配方,同时对比玻璃瓶与耐高压透明袋在多花黄精集约化生产中的应用效果。结果表明:多花黄精最佳外植体为成熟种子,种子在MS+6-BA2 mg·L−1+GA30.30 mg·L−1+NAA0.05 mg·L−1+蔗糖30 g·L−1的培养基中诱导率最高;在MS+6-BA 1.6 mg·L−1+清菌易0.07 g·L−1+蔗糖30 g·L−1的培养基中增殖效果较好,增殖系数达4.0以上;在MS+ NAA0.3 mg·L−1 +IBA0.6 mg·L−1 +蔗糖30 g·L−1的生根培养基中生根效果较好,生根率为95%。同时试验表明集约化生产多花黄精种苗整个生长周期均可用袋式培养代替瓶式培养。

       

      Abstract: In order to optimize the tissue culture and rapid propagation technology system of Polygonatum cyrtonema Hua, 70% mature berries, mature seeds, and the underground tubers of three-year-old seedlings of Polygonatum cyrtonema Hua were used as the experimental materials to screen the best explants. The further experiments were conducted to select the optimal induction medium, bud propagation subculture medium, and rooting medium for the best explants. At the same time, the application effects of glass bottles and high-pressure resistant transparent bags were compared in the intensive production of Polygonatum cyrtonema Hua. The results showed that the optimal explant of Polygonatum cyrtonema Hua was mature seeds, which had the highest induction rate in the medium of MS+6-BA2 mg·L−1+GA30.30 mg·L−1+NAA0.05 mg·L−1+sucrose 30 g·L−1. The proliferation effect was better in the medium of MS+6-BA 1.6 mg·L−1+Qingjunyi 0.07 g·L−1+sucrose 30 g·L−1, with the proliferation coefficient of over 4. The rooting effect was better in the rooting medium of MS+NAA0.3 mg·L−1+IBA0.6 mg·L−1+sucrose 30 g·L−1, with the rooting rate of 95%. At the same time, the experiments had shown that the bag-type tissue culture could be used instead of the bottle-type tissue culture for the entire growth cycle of intensive production of Polygonatum cyrtonema Hua seedlings.
