    Rapid Determination of 1091 Pesticide Residues in Edible Fungi by Liquid Chromatography Coupled with Quadrupole Time-of-flight Mass Spectrometry

    • 摘要: 为进一步增强食用菌中风险物质的非靶向筛查效率,基于LC-QTOF/MS分析技术,结合QuEChERS前处理,通过获取目标物的一级精确质量数和二级质谱碎片信息,建立了食用菌中农药及代谢物的高通量快速筛查平台。该方法以母离子质量偏差值≤5×10−6、同位素丰度差异≤5%、二级谱库匹配度≥70%为定性判定依据,无需标准物质,即可实现食用菌中1 091种农药及代谢物的快速筛查,16种代表性农药方法学验证表明,农药的检出限为2~1 000 ng·mL−1。该方法具有覆盖农药种类广、前处理简便、无需标准品、分辨率高且通量高等优势,适用于食用菌中广泛靶向农药残留的日常筛查,可为食品安全监管和风险评估提供可靠的技术支持。

       

      Abstract: In order to further enhance the efficiency of untargeted screening for hazardous substances in edible fungi, based on liquid chromatography coupled with quadrupole time-of-flight mass spectrometry (LC-QTOF/MS) combined with QuEChERS pretreatment, a high-throughput rapid screening platform for pesticides and their metabolites in edible fungi was established by acquiring the first-order accurate precursor mass values and secondary fragment spectral information of target analytes. The qualitative identification criteria were defined for the platform, including a precursor ion mass deviation of ≤5×106, an isotopic abundance difference of ≤5%, and a secondary spectral library matching score of ≥70%. This approach enabled the rapid screening of 1091 pesticides and their metabolites in edible fungi without the need for reference standards. The rapid screening of 1 091 pesticides and metabolites in edible fungi could be realized. The methodological validation by using 16 representative pesticides showed that the platform’s limits of detection (LODs) ranged from 2 to 1000 ng·mL−1. This method had the advantages of covering a wide range of pesticides, simple pretreatment procedures, freedom from reference standard dependence, as well as high resolution and high throughput, which was suitable for the routine broad-spectrum screening of pesticide residues in edible fungi, and could provide robust technical support for food safety supervision and risk assessment.
