喂食蛋黄油对DSS诱导小鼠急性溃疡性结肠炎的干预效果

    Intervention Effect of Egg Yolk Oil Feeding on DSS-Induced Acute Ulcerative Colitis in Mice

    • 摘要: 为探究蛋黄油对葡聚糖硫酸钠(DSS)诱导小鼠急性溃疡性结肠炎(UC)的改善作用,以ICR小鼠为模型,随机分为正常对照组、DSS模型组、蛋黄油低剂量组(2.5 mL·kg−1)和高剂量组(5.0 mL·kg−1),进行35 d蛋黄油喂食及7 d DSS诱导UC试验,测定小鼠体质量、疾病活动指数(DAI)、结肠长度、脏器指数、血清炎症因子(TNF-α、IL-6、IL-1β、IL-10)及总胆汁酸(TBA)含量,并观察结肠组织病理学变化。结果表明:与DSS模型组相比,蛋黄油喂食组小鼠体质量下降减缓,DAI评分降低,结肠长度增加(P<0.05),盲肠、脾脏、肝脏肿胀指数下降(P<0.05);血清促炎因子TNF-α、IL-6、IL-1β水平降低(P<0.05),抑炎因子IL-10水平升高(P<0.05);血清TBA含量降低(P<0.05);结肠病理损伤明显改善。蛋黄油高剂量组效果优于低剂量组。综上,蛋黄油对DSS诱导的小鼠急性UC具有改善作用,其机制与调控炎症因子平衡、改善胆汁酸代谢及修复肠道屏障有关,在高品质蛋黄油功能性食品开发中具有良好的应用前景。

       

      Abstract: To investigate the ameliorative effects of egg yolk oil on dextran sulfate sodium(DSS)-induced acute ulcerative colitis(UC)in mice, ICR mice were randomly divided into normal control group, DSS model group, egg yolk oil low-dose group(2.5 mL·kg1), and high-dose group(5.0 mL·kg1). The mice were fed with egg yolk oil for 35 days followed by 7 days of DSS induction for UC. Body weight, disease activity index(DAI), colon length, organ indices, serum levels of inflammatory cytokines(TNF-α, IL-6, IL-1β, IL-10)and total bile acids(TBA)were measured, and colonic histopathological changes were observed. The results showed that compared with the DSS model group, the egg yolk oil-fed groups exhibited attenuated body weight loss, reduced DAI scores(P<0.05), increased colon length(P<0.05), and decreased swelling indices of the cecum, spleen, and liver(P<0.05). Serum levels of pro-inflammatory cytokines TNF-α, IL-6, and IL-1β were decreased(P<0.05), while the level of anti-inflammatory cytokine IL-10 was increased(P<0.05); serum TBA content was also reduced(P<0.05). Colonic pathological damage was markedly ameliorated. The high-dose group showed better effects than the low-dose group. In conclusion, egg yolk oil exerts ameliorative effects on DSS-induced acute UC in mice, and the underlying mechanism may involve regulation of inflammatory cytokine balance, improvement of bile acid metabolism, and repair of intestinal barrier function. These findings suggest a promising application prospect for egg yolk oil in the development of high-quality functional foods.
