    水稻OsTZF3基因敲除和过量表达载体的构建与遗传转化

    Construction and Genetic Transformation of OsTZF3 Gene Knockout and Overexpression Vectors in Rice

    • 摘要: 为进一步研究水稻OsTZF3基因的生物学功能,利用日本晴基因组的OsTZF3基因序列,构建了OsTZF3的CRISP/Cas9敲除表达载体pKTZF3和过量表达载体pCXUN-TZF3;并通过农杆菌介导法导入粳稻日本晴胚性愈伤组织中,分别获得了pKTZF3和pCXUN-TZF3转化再生植株55个和42个克隆;进一步通过PCR法鉴定了35个克隆转pKTZF3植株和10个克隆转pCXUN-TZF3植株。

       

      Abstract: In order to further study the biological function of OsTZF3 gene in rice, the CRISP/Cas9 knockout expression vector pKTZF3 and overexpression vector pCXUN-TZF3 of OsTZF3 were constructed by using the sequence of OsTZF3 gene in Nipponbare. 55 transformants of pKTZF3 and 42 clones of pCXUN-TZF3 were obtained through Agrobacterium-mediated transformation. Among them, 35 clones of pKTZF3 and 10 clones of pCXUN-TZF3 were further identified by PCR detection.
