    响应面法优化绣球菌副产物多糖提取工艺的研究

    Study on Optimization of Extraction Process of Polysaccharides from Byproducts of Sparassis Crispa by Using Response Surface Method

    • 摘要: 为开发利用绣球菌副产物,提高绣球附加值,考察提取温度、提取时间、料水比3个因素对绣球菌副产物多糖得率的影响,在此基础上建立响应面分析模型,经线性回归拟合,回归模型:多糖得率(%)=7.19+0.26A-0.35B-0.84C-0.088AB-0.57AC+0.54BC-0.35A2-3.38B2-0.42C2P<0.000 1,达极显著水平;失拟项P=0.510 1,不显著,说明该模型成立。利用Design-Expert8.0软件对模型进行优化,以多糖得率最高为优化目标,得出绣球菌副产物多糖提取的最优工艺条件为:提取时间3.83 h,提取温度为78.75℃,料液比为1:30,多糖得率达到7.89%。根据实际情况将工艺参数修正为:提取时间3.83 h,提取温度为79.00℃,料液比为1:30。采用上述最优工艺条件进行绣球菌副产物多糖提取,测得实际多糖得率为8.01%。

       

      Abstract: In order to develop and utilize the byproducts of Sparassis crispa and increase the added value of hydrangea, the effects of extraction temperature, extraction time and ratio of feed to water on the polysaccharide yield of byproducts of Sparassis crispa were investigated. On this basis, the model of response surface analysiswas established, and by linear regression fitting, the regression model was obtained:polysaccharide yield (%)=7.19+0.26A-0.35B-0.84C-0.088AB-0.57AC+0.54BC-0.35A2-3.38B2-0.42C2, P<0.0001, reaching a very significant level. The lack of fit (P=0.5101) was not significant, indicating that the model was valid. Then, the model was optimized by using Design-Expert8.0 software, with the highest polysaccharide yield as the optimization target, and the optimal technological conditions for extracting polysaccharides from the byproducts of Sparassis crispa were as follows:extraction time was 3.83 h, extraction temperature was 78.75℃, and the solid-liquid ratio 1:30, thus the polysaccharide yield could be up to 7.89%. According to the actual situation, the technological parameters were revised as follows:extraction time was 3.83 h, extraction temperature was 79.00℃, and the solid-liquid ratio was 1:30. The polysaccharides were extracted from the byproducts of Sparassis crispa under the optimal conditions, and the actual polysaccharide yield was 8.01%.
