    酶解工艺对海带酶解液抑制血管紧张素转化酶活性的影响

    Effect of Enzymolysis Technology on the Inhibition of Angiotensin Converting Enzyme Activity by Kelp Enzyme Hydrolysate

    • 摘要: 为明确海带对血管紧张素转化酶活性的抑制作用,通过单因素试验和响应面优化试验,探讨不同纤维素酶添加量和酶解时间对海带浆抑制血管紧张素转化酶活性的影响。结果表明:采用纤维素酶对海带浆进行酶解,建立了海带酶解液血管紧张素转化酶(ACE)活性抑制率Y与纤维素酶添加量X1、酶解时间X2的数学模型:Y=-114.273 04+0.005 72X1+1.491 99X2-3.333 33×10-7X1X2-2.552 00×10-7X12-0.004 47X22,得到优化酶解条件为:海带经切分匀浆后,在pH 6.0、添加纤维素酶11 060 U·g-1、在室温条件下酶解169 min,所得酶解液对ACE活性的抑制率可达到41.43%左右。试验可知海带酶解液对血管紧张素转化酶(ACE)活性具有较高的抑制作用。

       

      Abstract: In order to clarify the inhibiting effect of kelp on the activity of angiotensin converting enzyme, the effects of different additive amount of cellulase and enzymolysis time on the inhibition of angiotensin converting enzyme activity by kelp pulp were investigated by the single factor experiment and response surface optimization test. The results showed that the enzymatic hydrolysis of kelp pulp with cellulase was carried out, and the mathematical model of the inhibition rate Y of ACE activity in kelp enzymatic hydrolysate, the enzyme additive amount X1 and the enzymolysis time X2 was established:Y=-114.273 04+0.005 72X1+1.491 99X2-3.333 33×10-7X1X2-2.552 00×10-7X12-0.004 47X22. The optimal enzymolysis condition was obtained as follows:after the kelp was cut and homogenized, 11 060 U·g-1 cellulase was added at pH 6.0 and the enzymatic hydrolysis was performed at room temperature for 169 min. The inhibition rate of the obtained enzymatic hydrolysate on ACE activity was about 41.43%. The results showed that the enzyme hydrolysate of kelp had a high inhibitory effect on the activity of angiotensin converting enzyme.
