    薄皮甜瓜嫁接砧穗愈合过程组织学观察

    Histological Observation on the Healing Process of Grafted Rootstock and Scion of Thin Skinned Cucumis Melo

    • 摘要: 为了解薄皮甜瓜嫁接后的砧穗亲和力和砧穗愈合情况,以广西当地主栽品种广蜜1号为接穗,选择广西农业科学院园艺研究所选育的白瓜砧BX-1、南瓜砧NX-3、南瓜砧NX-4等3个品系作为砧木,进行了嫁接后第2、4、6、8、10 d的砧穗愈合组织学观察。结果表明:白瓜砧BX-1、南瓜砧NX-3、南瓜砧NX-4均表现出良好的嫁接亲和力,嫁接后砧穗愈合部位的组织学变化及愈合速度基本相同。嫁接后第2 d,砧木和接穗各自形成一层隔离层,嫁接体处于隔离层形成阶段;嫁接后第4 d,隔离层颜色变浅,隔离层的边缘形成愈伤组织,嫁接体处于愈伤组织形成阶段;嫁接后第6 d,隔离层染色颜色明显变淡,愈伤组织进一步分化,形成愈伤组织桥;嫁接后第8 d,砧木和接穗切口相接处形成许多间断的愈伤组织桥,隔离层外侧的大部分空隙被愈伤组织填充;嫁接后第10 d,砧木和接穗切口相接处的隔离层几乎完全消失。

       

      Abstract: In order to understand the rootstock-scion compatibility and healing of thin skinned Cucumis melo after grafting, Guangmi No.1, the main local cultivar in Guangxi, was selected as the scion, and the three cultivars, white melon rootstock BX-1, pumpkin rootstock NX-3 and pumpkin rootstock NX-4, which were bred by Horticulture Institute of Guangxi Academy of Agricultural Science, were selected as the rootstocks. Then, the histological observation of the rootstock-scion healing on the 2nd, 4th, 6th, 8th and 10th days after grafting was carried out. The results showed that white melon rootstock BX-1, pumpkin rootstock NX-3 and pumpkin rootstock NX-4 all showed good grafting compatibility, and the histological changes of the rootstock-scion healing site and the healing speed after grafting were basically the same. On the 2nd day after grafting, the rootstock and scion formed an isolation layer respectively, and the graft was in the formation stage of isolation layer. On the 4th day after grafting, the color of the isolation layer became lighter, and the callus formed at the edge of the isolation layer, and the graft was in the formation stage of callus. On the 6th day after grafting, the color of the isolation layer became lighter obviously, and the callus differentiated further, forming callus bridges. On the 8th day after grafting, many discontinuous callus bridges were formed at the junction between the incisions of rootstock and scion, and most of the gap outside the isolation layer was filled with callus. On the 10th day after grafting, the isolation layer at the junction of rootstock-scion incision almost disappeared.
