    PurB过表达对大肠杆菌氨基糖苷类抗生素耐药性的影响

    Effect of PurB Overexpression on the Drug Resistance of Aminoglycoside Antibiotics in Escherichia coli

    • 摘要: 为寻找新的耐药靶点以解决细菌耐药问题,通过用pCA24N表达载体构建大肠杆菌腺苷酸琥珀酸裂解酶PurB过表达株(BW25113-purB),与对数期大肠杆菌携带空质粒(BW25113-pCA24N)进行氨基糖苷类抗生素杀伤效果比较,用ANOVA算法分析试验结果差异性,测试大肠杆菌PurB过表达在氨基糖苷类抗生素环境中的作用。结果显示PurB过表达后能够显著减弱庆大霉素以及卡那霉素对大肠杆菌的杀伤效果,证明PurB过表达影响了大肠杆菌对氨基糖苷类抗生素的耐受性。表明腺苷酸琥珀酸裂解酶在耐药大肠杆菌产生的进程中具有重要的作用,提示该酶作为大肠杆菌新的药理靶点的可行性。

       

      Abstract: In order to find a new drug resistance target to solve the problem of bacterial drug resistance, the PurB overexpression strain (BW25113-purB) of Escherichia coli adenylosuccinate lyase was constructed by using the pCA24N expression vector, and the killing effect of aminoglycoside antibiotics was compared with that of Escherichia coli carrying empty plasmid at the logarithmic phase (BW25113-pCA24N). Then, the ANOVA algorithm was used to analyze the difference of the test results, and the effect of PurB overexpression of Escherichia coli in the environment of aminoglycoside antibiotics was tested. The results showed that PurB overexpression could significantly attenuate the killing effect of gentamicin and kanamycin on Escherichia coli, proving that PurB overexpression affected the tolerance of Escherichia coli to aminoglycoside antibiotics. These results indicated that the adenylosuccinate lyase played an important role in the production process of drug-resistant Escherichia coli, suggesting the feasibility of this enzyme as a new pharmacological target for Escherichia coli.
