    基于ISSR分子标记的44种山药种质资源遗传多样性分析

    Analysis of the Genetic Diversity of 44 Yam Germplasm Resources Based on ISSR Molecular Markers

    • 摘要: 为探讨不同地区山药种质资源类型及遗传多样性亲缘关系,为山药品种选育、种质资源的保护利用提供参考。采用ISSR标记技术对44份山药种质资源进行遗传多样性及聚类分析。结果表明:从100条随机引物中筛选出8条随机引物,8条引物扩增获得的总谱带数为87条,多态性谱带数为87条,多态性谱带比例为100%。聚类分析将44份种质资源分成4个类群,从结果看ISSR标记技术聚类分析与植物分类学进行的基原分类结果基本一致,44份山药种质资源的遗传距离在0.0313~0.7216,以遗传距离为0.5618进行划分,可以将44份山药种质资源划分为5类,其中薯蓣与褐苞薯蓣亲缘关系最近,与山薯较远,与参薯距离最远。

       

      Abstract: In order to explore the types of yam germplasm resources and the genetic relationship of their genetic diversity in different regions, and provide reference for the breeding of Chinese yam varieties and the protection and utilization of germplasm resources, the genetic diversity and cluster analysis of 44 germplasm resources of Chinese yam were carried out by ISSR marker technique. The results showed that the 8 arbitrary primers were screened from 100 arbitrary primers, and the total number of bands amplified by 8 primers was 87, the number of polymorphic bands was 87, and the proportion of polymorphic bands was 100%. The cluster analysis divided the 44 germplasm resources into four class groups. The results of the cluster analysis by ISSR marker technique were basically consistent with the original classification results of plant taxonomy. The genetic distance of the 44 yam germplasm resources ranged from 0.0313 to 0.7216. According to the genetic distance of 0.5618, the 44 germplasm resources of yam could be divided into 5 categories. Among them, the genetic relationship between Dioscorea opposita and Dioscorea persimilis was the closest, which was far from Dioscorea opposita and the farthest from Dioscorea opposita.
