    产脂肪酶菌株的筛选及鉴定

    Screening and Identification of Lipase-producing Strains

    • 摘要: 为了挖掘更多的产脂肪酶菌种资源,通过橄榄油平板检测(透明圈观察),采用高通量从海水、滩涂泥壤以及滨海植物分离筛选了324株真菌菌株,在pH 9.4的平板上筛选获得了48个候选产脂肪酶菌株;经ITS序列分析,初步鉴定了其中的12个菌株,分别为镰刀菌、青霉属、枝孢菌、交链孢属等,其中多数为枝孢属和青霉属;进一步利用氢氧化钠滴定法检测其中2个菌株(#89和#260)培养24 h的发酵液,#89菌株产生的脂肪酶酶活为(6.92±2.40)U·mL-1,#260菌株产生的脂肪酶酶活为(7.67±1.70)U·mL-1

       

      Abstract: In order to explore more lipase-producing strain resources, 324 fungal strains were isolated and screened from the plants in seawater, coastal wetland & mud soil and coastal areas by high throughput through the olive oil plate detection (transparent circle observation). Forty-eight candidate lipase-producing strains were screened on the plate with pH 9.4. Through the ITS sequence analysis, 12 strains were preliminarily identified as Fusarium, Penicillium, Cladosporium, Alternaria, etc., most of which were Cladosporium and Penicillium. The fermentation broth of the two strains (#89 and #260) cultured for 24 h was further detected by using the titration method of sodium hydroxide. The lipase activity produced by #89 strain was (6.92±2.40) U·mL-1, and the lipase activity produced by #260 strain was (7.67±1.70) U·mL-1.
