Acute Toxicity Test of Compound Preparation of Jinlianju on Mice

Abstract
By taking the healthy Kunming mice as the animal model, the compound preparation of Jinlianju was given different doses (10.00, 7.76, 6.03, 4.68, 3.63 and 2.82 g·kg-1) by oral administration at one time. The acute toxicity test of mice was carried out to evaluate the drug safety. The results showed that after filling different doses of the compound preparation of Jinlianju, the mice showed quiet, laying still and abdominal breathing at first; As time went on, some mice showed clothing hair erection, protruding eyes and blue blood vessels in the tail. In the severe cases, the abdomen contracted sharply, the movement was unstable, and the mice twitched and died, but the toxic reaction of the mice basically disappeared at the 4th day after medication. The death of mice mainly occurred within 24 h after medication, and showed an obvious dose-effect relationship. The dead mice were found to have congestion in the lungs or bleeding points, as well as pleural effusion. After the end of the test, all the remaining mice were sacrificed for autopsy, and it was found that the internal organs of the mice in each treatment group had no obvious pathological changes, which was consistent with the internal organs of normal mice. By using the Karber method, the LD50 and Sx50 of Jinlianju compound preparation for mice were obtained, being 6.299 g·kg-1 and 0.035 5, respectively. And the confidence interval of 95% was 7.394-5.367, indicating that the compound preparation of Jinlianju belonged to the actual non-toxic substance.
