    Study on the Technologies of Tissue Culture and Rapid Propagation of Rubus Chingii

    • The optimal medium formula was selected for the tissue culture and rapid propagation of Rubus chingii. In this paper, the fine NS8 annual branch of Rubus chingii was used as the explant to conduct the tissue culture experiments under different hormone composition and concentration levels. The results showed that the best induction medium was MS+6-BA 1.5 mg·L-1+NAA 0.15 mg·L-1+GA3 0 mg·L-1, in which the induction rate reached 68%, and the bud seedlings were haleness. The best medium for proliferation was MS+6-BA 1.5 mg·L-1+NAA 0.1 mg·L-1, in which the growth state of the seedlings was haleness and the proliferation coefficient reached 4.87. The best medium for rooting was 1/2MS+NAA 0.6 mg·L-1+IBA 0.2 mg·L-1+ activated carbon 0.8 g·L-1, in which the rooting rate reached 95.3%, the number of rooting was 5.7, and the root length was 5.8 cm, with well developed root system and good growth vigor of seedlings. The results showed that the selected best medium formula was suitable for the tissue culture and rapid propagation of local Rubus chingii, which would provide the basis for obtaining a large number of high-quality seedlings and large-scale commercial production in a short period of time.
