Identification and Domestication Cultivation of a Wild Ganoderma lucidum Plant in Huangcuiwei Mountain of Shanghang

Abstract
The pure strain QS21-4 was isolated from the fruiting body tissue of a wild Ganoderma lucidum plant in Huangcuiwei Mountain of Shanghang. And the strain QS21-4 was identified as Ganoderma sinense based on the morphological characteristics of fruiting bodies and ITS sequencing results. Then, the extraction of effective active ingredients from the mycelia of the strain QS21-4 by liquid fermentation, the screening of different pathogen culture medium formulas, and the detection of effective active ingredients in the fruiting bodies cultivated under forests were carried out. The results showed that the formula B (broadleaved sawdust 78%, wheat bran 15%, corn cracked 5%, sucrose 1%, gypsum powder 1%) was the most suitable for the mycelial growth of the strain QS21-4. The contents of polysaccharides and triterpenoids in the mycelia of the strain QS21-4 by liquid fermentation and the fruiting bodies of the strain cultivated under forests were higher than those of the control strain Wuzhi No.2. The stipe of fruiting body of the strain QS21-4 cultivated under forests was shorter than that of the control strain Wuzhi No.2, while the pileus diameter and fresh weight per bag of Ganoderma lucidum were similar to those of the control strain.
