Analysis on the Enhancement of Aminoglycoside and Quinolone Antibiotics’ Effect on Killing Escherichia Coli by Coenzyme Q0

Abstract
Finding the new-type antibiotic adjuvant to improve the bactericidal efficiency of the existing antibiotics and achieve the rapid and efficient killing of pathogens is an important means to reduce the risk of bacterial resistance. The bactericidal effect of coenzyme Q0 (CoQ0) combined with aminoglycosides or quinolones antibiotics against Escherichia coli was tested by adding the exogenous coenzyme Q0. The results showed that the coenzyme Q0 could significantly enhance the bactericidal effect of aminoglycosides and quinolones antibiotics against Escherichia coli BW25113 and 8 strains of clinical drug-resistant Escherichia coli. The results of concentration gradient test and time-dependent test showed that the coenzyme Q0 enhanced the bactericidal activity of aminoglycosides and quinolones antibiotics in a concentration-dependent and time-dependent manner. The optimal concentration of coenzyme Q0 was 30 µg·mL−1. The optimal action time of coenzyme Q0 assisted gentamicin was 5 h, and the optimal action time of coenzyme Q0 assisted ofloxacin was 8 h. The results provided data support for the development of coenzyme Q0 as a new-type adjuvant for aminoglycoside or quinolones antibiotics.
