Molecular identification of hybrid progeny in Phalaenopsis

Abstract
To verify the authenticity of hybrid species in Phalaenopsis and screen for true hybrids with superior parental traits. This study focused on two parents of Phalaenopsis, 'Qiaoyiailisi' and 'Yixiantian', as well as their 138 F1 hybrid progeny. Three pairs of SRAP primers were selected from 50 pairs for the identification of true and false hybrids in hybrid progeny. The results showed that three pairs of SRAP primers me2-em16, me4-em47, and me1-em9 could classify the amplification band types of the hybrid population into true hybrid type, maternal type, paternal type, and deletion type. Furthermore, 135 true hybrids were identified from 138 F1 hybrid populations, accounting for 97.8% of the hybrid population. Therefore, using SRAP molecular marker technology to identify hybrid populations of Phalaenopsis is feasible, and the relevant results lay the foundation for the construction of genetic maps and QTL mapping of Phalaenopsis.
